Human Persephin (PSPN) ELISA Kit

A sandwich ELISA from Assay Genie for the precise measurement of persephin (PSPN) levels in human samples, this kit uses a colorimetric detection method and is compatible with serum, plasma, cell culture supernatant, cell or tissue lysates, and other liquid samples. With a sensitivity of 18.75 pg/mL and a detection range of 31.25–2000 pg/mL, it is designed to support research into persephin's role in promoting neuron survival and development, including investigations relevant to neurological conditions.

The assay procedure includes reagent and plate preparation, primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody binding at 37°C for 60 minutes, HRP-Streptavidin (SABC) binding at 37°C for 30 minutes, TMB substrate colour development in the dark for 10–20 minutes, and absorbance reading at 450 nm after stop solution addition. Sample preparation protocols are provided for serum (centrifuge at 1000 × g for 20 minutes), plasma (centrifuge at 1000 × g for 15 minutes at 2–8°C), tissue homogenate (in PBS with protease inhibitors), cell culture supernatant (centrifuge at 2500 rpm for 5 minutes), and cell lysate.

Linearity has been evaluated across dilutions of 1:2, 1:4, and 1:8, with serum ranging 83–100%, 81–100%, and 85–103% respectively, EDTA plasma 85–100%, 82–95%, and 87–105%, and heparin plasma 84–95%, 86–100%, and 85–104%. Recovery averages are 93% for serum, 96% for EDTA plasma, and 96% for heparin plasma. The kit is stored at 2–8°C for 12 months.
