Periplakin is a plakin-family cytoskeletal linker expressed in stratified and simple epithelia, where it stabilizes desmosomal attachments, contributes to the cornified envelope of keratinocytes, and supports epidermal barrier integrity. Quantification of PPL protein levels in clinical and experimental samples helps researchers probe keratinization disorders, mucosal barrier function, and epithelial differentiation programs.

This colorimetric immunoassay from Assay Genie uses a matched antibody pair to capture human periplakin from serum samples, plasma samples, tissue homogenate preparations, and cell lysates, then reports signal via an enzyme-substrate readout on a microplate reader. The kit provides pre-coated plate wells, lyophilized reference standards, biotinylated detection antibody, streptavidin-HRP conjugate, wash buffer, substrate solution, and stop reagent in quantities sufficient for 96 individual reactions. Reconstituted standards and working-strength detection antibody should be prepared fresh; leftover aliquots can be frozen for repeat use within the kit's stated shelf life.

Applications include profiling PPL expression in lesional versus non-lesional skin biopsies, tracking barrier disruption in inflammatory dermatoses, evaluating periplakin involvement in epidermolysis bullosa simplex and related keratinization disorders, and characterizing epithelial-mesenchymal transition in mucosal tissue models. Store unopened kit components refrigerated and protected from light until first use; reconstituted working solutions are best held frozen in single-use aliquots to limit freeze-thaw cycles.
