Human Perforin ELISpot Kit

The Assay Genie Human Perforin ELISpot Kit enables single-cell detection of perforin secretion by cytotoxic T lymphocytes and natural killer cells. Perforin is a 67 kDa pore-forming effector protein stored in the granules of CD8+ T cells and NK cells, where it polymerizes on target membranes to deliver granzyme-mediated apoptosis.

This ELISpot format uses a PVDF-membrane microplate pre-coated with a human-specific capture antibody, allowing individual secreting cells to form discrete, countable spots after a short cell culture incubation. The plate is developed with biotinylated detection antibody, streptavidin-enzyme conjugate, and AEC substrate, and spots are enumerated on an automated reader or stereomicroscope, with each spot representing one perforin-releasing cell.

Reconstituted detection reagents should be aliquoted and stored refrigerated to preserve activity, while the substrate is protected from light. Working volume per well is 100 uL during cell incubation. For research use only, not for diagnostic procedures.
