Human Pepsin ELISA Kit

Pepsin is a key enzyme involved in the digestive process, breaking down proteins in the stomach, and abnormal pepsin levels can indicate certain gastrointestinal conditions and disorders, making it a critical biomarker for studying and diagnosing such issues.

The kit uses a colorimetric, double-antibody sandwich ELISA format with a sensitivity of 37.5 pg/mL and a detection range of 62.5-4000 pg/mL. It is reactive with human samples and validated for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reagents should be stored at 2-8°C for up to 12 months.

The assay procedure involves equilibrating reagents and TMB substrate to room temperature, loading standards and samples on the pre-coated plate, incubating at 37°C for 90 minutes for antigen binding, adding biotin-labeled detection antibody with a 60-minute incubation, adding HRP-Streptavidin (SABC) for 30 minutes, developing with TMB substrate in the dark for 10-20 minutes, and reading absorbance at 450 nm after adding stop solution. Sample preparation guidelines include centrifuging serum at 1000 × g for 20 minutes, plasma at 1000 × g for 15 minutes at 2-8°C, and cell culture supernatant at 2500 rpm for 5 minutes.
