Description

A sandwich ELISA from Assay Genie is designed for the quantitative measurement of PDK4 (Pyruvate Dehydrogenase Kinase Isozyme 4) in human samples. The assay uses a double-antibody sandwich format with colorimetric detection, and the signal is read as absorbance at 450 nm. It is reactive with human samples and has been validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit targets PDK4, a key regulator of the pyruvate dehydrogenase complex that plays a central role in energy metabolism and glucose utilization.

The reported sensitivity is 0.938 ng/mL and the working range is 1.563-100 ng/mL. Recovery has been demonstrated in serum (range 92-105%, average 97%), EDTA plasma (range 86-99%, average 93%), and heparin plasma (range 87-105%, average 93%). Linearity is supported by dilutional parallelism across 1:2, 1:4, and 1:8 dilutions: serum 85-99%, 82-96%, 82-95%; EDTA plasma 83-97%, 80-99%, 82-95%; heparin plasma 82-98%, 80-100%, 83-97% (n=5 each). Unopened reagents should be stored at 2-8°C for 12 months.

The workflow involves equilibration of reagents, a primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody incubation at 37°C for 60 minutes, HRP-Streptavidin (SABC) binding at 37°C for 30 minutes, TMB colour development in the dark for 10-20 minutes, and a stop step followed by immediate absorbance measurement at 450 nm. Intended for research in metabolism, the kit supports studies of PDK4 dysregulation in metabolic disorders such as diabetes, obesity, and mitochondrial conditions, providing a tool for biomarker investigation and therapeutic research.
