PDHX (Pyruvate Dehydrogenase Complex Component X), also called the E3-binding protein, anchors the E3 component within the mitochondrial pyruvate dehydrogenase complex, the enzyme gatekeeper that decarboxylates pyruvate into acetyl-CoA for entry into the TCA cycle. Recessive PDHX mutations cause pyruvate dehydrogenase deficiency with lactic acidosis, neurodevelopmental delay, and basal ganglia injury, and altered PDHX expression has been reported in tumor metabolism studies of the Warburg effect and in models of mitochondrial biogenesis and glucose oxidation.
