Human PDHA2 ELISA Kit

A Sandwich ELISA with double-antibody, colorimetric detection from Assay Genie, intended for the quantification of human PDHA2 (Pyruvate Dehydrogenase E1 Alpha 2 Subunit) in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. PDHA2 is a mitochondrial enzyme that converts pyruvate into acetyl-CoA, and its dysregulation has been linked to metabolic disorders including diabetes, Alzheimer's disease, and various cancers.

Linearity across 1:2, 1:4 and 1:8 dilutions was reported at 90-103%, 87-97%, 89-96% for serum; 87-98%, 86-100%, 86-96% for EDTA plasma; and 80-96%, 80-96%, 85-97% for heparin plasma (n=5 each). Recovery averaged 94% for serum (range 86-104%), 93% for EDTA plasma (85-104%), and 92% for heparin plasma (85-99%).

The assay follows a six-step sandwich protocol: equilibrate reagents and TMB substrate, then load standards, samples and controls for a 90-minute primary incubation at 37°C; add biotin-labeled detection antibody for 60 minutes; add HRP-Streptavidin for 30 minutes; develop with TMB substrate in the dark for 10-20 minutes; then add stop solution and read absorbance at 450 nm. The kit is intended for research in the metabolism area, with components stored at 2-8°C for 12 months.
