Human PDGF-AB/Platelet-Derived Growth Factor AB ELISA Kit

Designed for the accurate measurement of PDGF-AB levels, this Sandwich ELISA kit from Assay Genie uses a double-antibody format with colorimetric detection. It is reactive with human samples and validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples.

The assay has a stated sensitivity of 9.375 pg/mL and a measuring range of 15.625–1000 pg/mL. Linearity in serum (n=5) was 92–102% (1:2), 82–101% (1:4) and 80–100% (1:8); in EDTA plasma (n=5) 89–102% (1:2), 90–100% (1:4) and 82–93% (1:8); and in heparin plasma (n=5) 88–104% (1:2), 84–101% (1:4) and 81–98% (1:8). Spike-and-recovery values are 88–103% (average 93%) in serum, 92–105% (average 96%) in EDTA plasma, and 92–93% (average 92%) in heparin plasma. PDGF-AB is described as a key growth factor involved in the regulation of cell growth and division, with roles in wound healing, tissue repair and vascular remodelling, and its dysregulation has been linked to cancer, fibrotic disorders and cardiovascular disease.

The protocol involves equilibrating reagents and TMB substrate to room temperature, a primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody for 60 minutes, HRP-Streptavidin (SABC) for 30 minutes, TMB development in the dark for 10–20 minutes, and absorbance reading at 450 nm after addition of stop solution. The protocol supplied is a sample protocol specific to each batch/lot. Kit components are stored at 2–8°C for 12 months, with the microplate strips and lyophilized standard additionally suitable for 12 months at −20°C.
