Description

A Sandwich ELISA employing a double-antibody format from Assay Genie, designed for the precise quantification of human PDE7B (cAMP-specific 3',5'-cyclic phosphodiesterase 7B) in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The colorimetric assay supports research into the enzyme's role in cAMP regulation and its links to cancer, inflammatory conditions, and neurological disorders.

Quoted performance figures from the manufacturer include a sensitivity of 0.188 ng/mL across a working range of 0.313–20 ng/mL. Linearity across 1:2, 1:4 and 1:8 dilutions (n = 5 each): serum 92–104%, 85–98%, 89–100%; EDTA plasma 90–103%, 86–96%, 84–98%; heparin plasma 87–102%, 82–98%, 81–96%. Spike-and-recovery (n = 5 each): serum 87–103% (average 96%), EDTA plasma 88–104% (average 98%), heparin plasma 87–104% (average 96%).

The procedure comprises six steps: reagent and plate preparation; primary incubation at 37°C for 90 minutes; biotin-labelled detection antibody incubation at 37°C for 60 minutes; HRP-Streptavidin (SABC) binding at 37°C for 30 minutes; TMB colour development in the dark for 10–20 minutes; and stop solution with absorbance reading at 450 nm. Reagents are stored at 2–8°C for 12 months.
