Description

This Assay Genie sandwich ELISA kit enables quantitative detection of human programmed cell death protein 4 (PDCD4) using a double-antibody configuration with a colorimetric readout. It is validated for use with serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, and is positioned within the Cell Death research area. Annotated target synonyms include PDCD4, H731, nuclear antigen H731, neoplastic transformation inhibitor protein, programmed cell death 4, Protein 197/15a, and nuclear antigen H731-like (UniProt Q53EL6; KEGG K16865; Gene ID 27250).

The reported analytical sensitivity is 0.188 ng/mL with an assay range of 0.313–20 ng/mL. Linearity was assessed at 1:2, 1:4 and 1:8 dilutions: serum (n = 5) 98–104%, 91–104%, 89–102%; EDTA plasma (n = 5) 82–86%, 82–101%, 83–93%; heparin plasma (n = 5) 83–96%, 90–99%, 80–97%. Spike-and-recovery across the same matrices averaged 93% in serum (range 87–102%), 98% in EDTA plasma (range 88–102%) and 97% in heparin plasma (range 85–105%). Reagents should be stored at 2–8°C, where they remain stable for 12 months.

The workflow follows a standard sandwich ELISA: reagent and plate equilibration, primary incubation at 37°C for 90 minutes, biotinylated detection antibody at 37°C for 60 minutes, HRP-streptavidin (SABC) at 37°C for 30 minutes, TMB substrate development in the dark for 10–20 minutes, and absorbance reading at 450 nm after stop solution. Sample-specific guidance is given for serum (clot, 1000 × g, 20 minutes), plasma (anticoagulant tubes, 1000 × g, 15 minutes, 2–8°C), tissue homogenates, cell culture supernatant (2500 rpm, 5 minutes) and cell lysates. PDCD4 is described as a regulator of cell growth, differentiation and apoptosis whose dysregulation has been linked to cancer, inflammatory disorders and neurological conditions.
