Human PDCD1LG2 (Programmed Cell Death Protein 1 Ligand 2) ELISA Kit (HUFI04749)

A colorimetric sandwich ELISA from Assay Genie designed for the quantitative detection of human PDCD1LG2, also known as PD-L2, CD273 or B7-DC. The kit accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, with a reported sensitivity of 28.125 pg/mL and an assay range of 46.875–3000 pg/mL. Reagents are stored at 2–8°C and remain stable for up to 12 months. PDCD1LG2 is a ligand involved in regulating immune responses and T cell activation, and its dysregulation has been implicated in cancer and autoimmune disorders, making the kit a useful tool for studies of immune regulation and potential therapeutic targets.

The double-antibody sandwich protocol is run on a pre-coated microplate. Standards, samples and controls are incubated at 37°C for 90 minutes, followed by a biotinylated detection antibody (60 minutes at 37°C) and HRP-Streptavidin conjugate (30 minutes at 37°C). TMB substrate is added for 10–20 minutes in the dark, then the reaction is stopped and absorbance is read at 450 nm. Reported linearity for serum, EDTA plasma and heparin plasma covers 1:2, 1:4 and 1:8 dilutions. Recovery ranges are 87–105% (serum, average 95%), 89–104% (EDTA plasma, average 96%) and 87–99% (heparin plasma, average 94%).

Sample preparation guidance covers serum (centrifuge at 1000 × g for 20 minutes after clotting), plasma (anticoagulant tubes, 1000 × g, 15 minutes at 2–8°C), tissue homogenate in PBS with protease inhibitors, cell culture supernatant (2500 rpm, 5 minutes) and cell lysate.
