Description

This sandwich ELISA from Assay Genie is designed for the quantitative measurement of human PDCD1 (Programmed Cell Death 1; also known as PD-1 and CD279; UniProt Q15116) in human samples. It uses a double antibody detection format with a colorimetric readout measured at 450 nm, and is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid sample types.

Reported performance characteristics include a sensitivity of 18.75 pg/mL and a working range of 31.25-2000 pg/mL. Spiked recovery is reported as 85-102% (average 93%) in serum (n=5), 92-105% (average 97%) in EDTA plasma (n=5), and 89-104% (average 94%) in heparin plasma (n=5). Dilutional linearity at 1:2, 1:4 and 1:8 is reported as 85-100%, 92-104% and 87-104% in serum; 87-94%, 82-99% and 82-96% in EDTA plasma; and 84-100%, 80-93% and 90-100% in heparin plasma. Unopened kit components are stored at 2-8°C for 12 months.

The assay workflow comprises a primary incubation at 37°C for 90 minutes, a biotin-labelled detection antibody step at 37°C for 60 minutes, HRP-Streptavidin binding at 37°C for 30 minutes, TMB colour development in the dark for 10-20 minutes, and a stop step before absorbance is read. According to the source material, PDCD1 is an immune checkpoint protein described as a regulator of immune responses and immune tolerance, with dysregulation linked to cancer, autoimmune disorders and infectious diseases.
