Paxillin is a ~68 kDa focal adhesion phosphoprotein that localizes to integrin-mediated adhesion sites, where it links the cytoskeleton to the extracellular matrix and is phosphorylated on tyrosine and serine residues during cell adhesion, spreading, and migration. Changes in paxillin abundance and phosphorylation status are widely used as read-outs of focal adhesion signaling in studies of cell motility, mechanotransduction, and tumor invasion, making quantitative immunoassay measurement of the human protein useful in cancer research and cytoskeletal signaling workflows. Each kit from Assay Genie is configured as a 96-assay immunoassay with a pre-coated microplate, biotinylated detection reagent, standard, streptavidin-enzyme conjugate, wash buffer, substrate, and stop solution, supplied in sealed foil pouches and individual vials.

Tissue homogenates prepared in cold lysis buffer with protease and phosphatase inhibitors are the validated sample matrix, since preserving phosphorylation state is critical for downstream signaling studies. Reconstitute the standard and store the reconstituted stock frozen in single-use aliquots to limit freeze-thaw cycles; refrigerate unopened components, protect the substrate from light, and bring all reagents to room temperature before running the assay. This product is intended for research use only and is not for diagnostic procedures.

For research applications in cell biology, ELISA-based quantification of paxillin is often paired with western blot and immunoprecipitation to confirm phosphorylation at the well-characterized Tyr31 and Tyr118 sites, or used alongside immunohistochemistry on adherent cell populations. Researchers studying focal adhesion dynamics, integrin signaling, or migration phenotypes in tumor and stromal cell models will find this kit integrates into existing cell culture and adhesion assay workflows.
