Paraoxonase 1 (PON1) is a calcium-dependent arylesterase synthesized in the liver and secreted into circulation, where it associates with high-density lipoprotein (HDL) and hydrolyzes oxidized lipid substrates, contributing to the antioxidant defense of serum. Quantitative measurement of human PON1 protein supports research in cardiovascular biology, metabolic disease, and toxicology, since altered PON1 expression has been linked to atherosclerosis, type 2 diabetes, hepatic dysfunction, and occupational organophosphate exposure. The Assay Genie kit includes a pre-coated microplate, biotinylated detection antibody, streptavidin-HRP conjugate, calibrated protein standards, wash buffer, and substrate, sufficient for 96 reactions. Typical sample volume is 20 µL of serum or plasma per well, with the full procedure run over a single working day at room temperature (18 to 25 degrees C). Unopened components should be refrigerated; reconstituted standards should be aliquoted and stored frozen to limit freeze-thaw degradation. Reagents remain stable for approximately 6 months from manufacture when handled as directed. For research use only; not for clinical diagnostic use.
