Pappalysin-2 (PAPP-A2) is a zinc-dependent metalloproteinase that cleaves insulin-like growth factor binding proteins IGFBP-3 and IGFBP-5, releasing bioactive IGF-1 and IGF-2 and modulating local growth factor signaling. The enzyme is expressed in placental tissue, developing bone, and reproductive compartments, and altered circulating levels have been investigated as a biomarker for fetal growth trajectories, preeclampsia, and metabolic bone disorders.
