Description

Supplied by Assay Genie as a colorimetric Sandwich ELISA, this kit is designed for the quantification of human Pancreatic Secretory Trypsin Inhibitor (SPINK1) in serum, plasma, tissue homogenates, cell culture supernates and other biological fluids. The assay is reactive with human samples and detects natural and recombinant human Serine protease inhibitor Kazal-type 1. SPINK1 is also known by the synonyms Pancreatic secretory trypsin inhibitor, Tumor-associated trypsin inhibitor, TATI, and PSTI, and is catalogued at UniProt P00995 as a secreted protein.

Performance is reported as a detection range of 0.312-20ng/mL with a sensitivity of 0.13ng/mL. Intra-assay CV is 4.8% and inter-assay CV is 8.0%. Dilutional linearity in serum (N=5) is 90-102% at 1:2, 87-96% at 1:4, 110-122% at 1:8, and 104-114% at 1:16; EDTA plasma (N=5) is 98-108%, 84-93%, 87-96%, and 84-95% across the same dilutions; heparin plasma (N=5) is 102-111%, 103-113%, 85-95%, and 99-109%. Recovery averages 101% (95-107%) for serum and 103% (97-109%) for plasma. The kit is for research use only, with the research area listed as cancer.

SPINK1 has an additional role in the male reproductive tract, where it binds to sperm heads and modulates sperm capacitation by inhibiting calcium uptake and nitrogen oxide (NO) production. Defects in SPINK1 are associated with hereditary pancreatitis and tropical calcific pancreatitis, supporting use in studies of pancreatic function, pancreatitis, and pancreatic cancer.
