Description

Manufactured by Assay Genie, this sandwich ELISA kit is designed for the quantitative measurement of human Pancreatic Lipase (PNLIP) using a double-antibody format. The assay is intended for research in the area of metabolism and is reactive with human samples. Pancreatic lipase is an essential enzyme involved in the digestion and absorption of dietary fats, and abnormal levels have been associated with pancreatic diseases, malnutrition, and obesity, making it a useful marker for studies of pancreatic health and lipid metabolism.

The kit reports a sensitivity of 2.813 mIU/mL and a measuring range of 4.688–300 mIU/mL. It is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Linearity for serum (n=10) is 87–104% at 1:2, 82–101% at 1:4, and 80–98% at 1:8; EDTA plasma (n=10) shows 90–103%, 89–100%, and 89–98%; and heparin plasma (n=10) shows 87–100%, 83–93%, and 83–99% across the same dilutions. Recovery ranges are 87–105% (serum, average 98%), 85–100% (EDTA plasma, average 94%), and 85–102% (heparin plasma, average 92%). Reagents should be stored at 2–8°C for 12 months.

The procedure involves equilibrating reagents and TMB substrate to room temperature before loading standards, samples and blanks for a 90-minute primary incubation at 37°C. A biotin-labelled detection antibody is then applied for 60 minutes, followed by HRP-Streptavidin conjugate for 30 minutes. Colour development with TMB substrate proceeds for 10–20 minutes in the dark, after which the reaction is stopped and absorbance is read at 450 nm.
