Human PADI4/PAD4 ELISA Kit

A sandwich, double-antibody ELISA from Assay Genie designed to quantify PADI4 (also known as PAD4) in human samples. PADI4 is a peptidylarginine deiminase enzyme involved in protein citrullination, with roles in gene regulation and immune response. Abnormal PADI4 levels have been linked to autoimmune diseases, inflammatory conditions and cancer, making it a biomarker of interest in immunology research.

The kit uses a colorimetric detection method with a reported sensitivity of 0.113 ng/mL and an assay range of 0.188-12 ng/mL. Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reported linearity across 1:2 to 1:8 dilutions ranges from 80% to 104% for serum, EDTA plasma and heparin plasma, with spike-recovery averages of 98% (serum), 94% (EDTA plasma) and 93% (heparin plasma). Kits should be stored at 2-8°C for up to 12 months.

The workflow involves equilibrating reagents, a primary incubation at 37°C for 90 minutes, biotinylated detection antibody binding at 37°C for 60 minutes, HRP-streptavidin binding for 30 minutes, TMB colour development in the dark for 10-20 minutes, and absorbance reading at 450 nm after adding stop solution.
