Human PACRG/Parkin coregulated gene protein ELISA Kit

From Assay Genie, this Sandwich ELISA (double antibody) kit is designed for the quantitative detection of human PACRG (Parkin coregulated gene protein) in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The colorimetric assay has a stated sensitivity of 0.188 ng/mL and a detection range of 0.313-20 ng/mL. Reagents are stored at 2-8°C for 12 months.

Validation data supplied by the manufacturer includes linearity across dilutions of 1:2, 1:4 and 1:8: serum 90-101% to 91-102%, EDTA plasma 84-100% to 89-105%, and heparin plasma 82-100% to 84-100%. Spike-and-recovery averages are 98% (serum, range 93-102%), 97% (EDTA plasma, range 85-104%) and 96% (heparin plasma, range 88-102%). The protocol runs as a sandwich ELISA: equilibrate reagents, incubate standards and samples at 37°C for 90 minutes, add biotin-labelled detection antibody for 60 minutes at 37°C, incubate with HRP-Streptavidin (SABC) for 30 minutes at 37°C, develop with TMB substrate in the dark for 10-20 minutes, then stop and read absorbance at 450 nm.

Sample preparation guidance is provided for serum (clot, centrifuge at 1000 × g for 20 minutes), plasma (centrifuge anticoagulated blood at 1000 × g for 15 minutes at 2-8°C), tissue homogenates (in PBS with protease inhibitors), cell culture supernatant (centrifuge at 2500 rpm for 5 minutes) and cell lysates (lysis buffer with protease inhibitors). The kit is supplied with an ELISA microplate, lyophilised standard, biotin-labelled antibody concentrate (100X), HRP-Streptavidin concentrate (SABC, 100X), TMB substrate, sample dilution buffer, antibody dilution buffer, SABC dilution buffer, stop solution, 25X wash buffer, plate sealer and a technical manual.
