P5CS (delta-1-pyrroline-5-carboxylate synthase) is the mitochondrial enzyme encoded by the ALDH18A1 gene that catalyzes the rate-limiting step of proline biosynthesis from glutamate. Measuring circulating P5CS concentrations supports research into proline and arginine metabolism, collagen biosynthesis, and connective tissue biology, including investigations of ALDH18A1 dysfunction in hereditary spastic paraplegia, autosomal recessive cutis laxa, and neurocutaneous syndromes.
