Human P2RY12/P2Y purinoceptor 12 ELISA Kit

A sandwich double-antibody assay from Assay Genie designed for the quantitative detection of human P2RY12 (P2Y purinoceptor 12) in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. P2RY12 is a purinergic receptor involved in platelet activation and aggregation and plays a central role in hemostasis and thrombosis. Dysregulation of the receptor has been linked to cardiovascular diseases and inflammatory disorders, supporting its relevance in cardiovascular research.

The protocol follows a standard sandwich ELISA workflow: equilibration of reagents and TMB substrate to room temperature, loading of standards, samples and zero controls onto the pre-coated plate, primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody incubation at 37°C for 60 minutes, HRP-Streptavidin binding at 37°C for 30 minutes, TMB colour development in the dark for 10–20 minutes, and absorbance reading at 450 nm after adding stop solution.

Validation data provided by the manufacturer show linearity ranges of 89–104% (serum, 1:2 dilution), 82–100% (EDTA plasma, 1:2) and 88–104% (heparin plasma, 1:2), with spike-and-recovery ranges of 89–103% for serum, 87–101% for EDTA plasma and 90–105% for heparin plasma. Sample-specific preparation protocols are described for serum, plasma, tissue homogenate, cell culture supernatant and cell lysate.
