Description

From Assay Genie, this Sandwich ELISA kit is designed for the quantitative measurement of osteocalcin in human samples, including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Osteocalcin is described as a key protein involved in bone metabolism and essential for maintaining healthy bone density and strength, supporting research into bone formation, remodeling, mineralization, osteoporosis, fractures, and related metabolic conditions.

The assay uses a colorimetric, double-antibody sandwich format with a sensitivity of 0.141 ng/mL and a working range of 0.234-15 ng/mL. Reported linearity across serum, EDTA plasma, and heparin plasma at dilutions of 1:2, 1:4, and 1:8 falls within 80-103%, while average recoveries are 95% (serum), 96% (EDTA plasma), and 95% (heparin plasma). The kit is intended for use with human samples only and is stored at 2-8°C for 12 months. The protein target corresponds to UniProt P02818 and is also known by synonyms including OC, BGLAP, BGP, OCN, bone gamma-carboxyglutamate, gla protein, Bone Gla protein, and Gamma-carboxyglutamic acid-containing protein.

The supplied protocol describes equilibration of reagents and TMB substrate, primary incubation at 37°C for 90 minutes, biotin-labeled detection antibody binding for 60 minutes, HRP-Streptavidin (SABC) incubation for 30 minutes, TMB color development in the dark for 10-20 minutes, and absorbance measurement at 450 nm after stop solution addition. The kit has been cited in published research, including Oley et al. on serum BMP-2 and osteocalcin levels post alveolar bone graft in cleft lip and palate patients (Heliyon, 2023) and Dziadek et al. on PCL/bioactive glass biomaterials as carriers for polyphenolic compounds (Bioactive Materials, 2021).
