Description

A colorimetric Sandwich ELISA from Assay Genie designed for the precise measurement of ornithine aminotransferase (OAT) levels in human serum, plasma, and cell culture supernatants, with additional support for cell or tissue lysates and other liquid samples. OAT is an essential enzyme involved in amino acid metabolism, catalysing the conversion of ornithine to glutamate within the urea cycle, and its activity is relevant to research into metabolic disorders and liver diseases. Detection is read spectrophotometrically at 450 nm following a sequential incubation protocol: primary antigen binding at 37°C for 90 minutes, biotin-labelled detection antibody for 60 minutes, HRP-Streptavidin (SABC) for 30 minutes, and TMB colour development in the dark for 10–20 minutes, terminated with stop solution.

The kit reports a sensitivity of 0.188 ng/mL and an assay range of 0.313-20 ng/mL, and is stored at 2-8°C for 12 months. Linearity in serum (n=5) spans 85-104% at 1:2, 92-97% at 1:4 and 89-105% at 1:8, with EDTA plasma at 92-97%, 90-101% and 83-100%, and heparin plasma at 83-96%, 83-94% and 84-100% across the same dilutions. Recovery averages are reported as 93% for serum, 91% for EDTA plasma and 96% for heparin plasma across the relevant recovery ranges. Sample preparation protocols cover serum, plasma, tissue homogenate, cell culture supernatant and cell lysate workflows.
