Human Orexin (OX) ELISA Kit

The Assay Genie Human Orexin ELISA enables quantitative measurement of orexin A (hypocretin-1), a 33-amino acid neuropeptide synthesized by neurons of the lateral hypothalamus. Orexin signaling regulates sleep-wake transitions, feeding behavior, reward processing, and energy homeostasis, and altered cerebrospinal fluid orexin levels serve as a biomarker for narcolepsy type 1 and cataplexy. Researchers also use this assay to study orexin involvement in addiction, metabolic syndrome, and stress responses. Each well requires 50 uL of sample or standard, and the protocol completes in roughly 2.5 hours with incubation at 37 °C. Unopened kit components should be refrigerated; reconstituted standards and the biotinylated detection antibody require freezing at -20 °C and should be aliquoted to limit freeze-thaw degradation. The kit includes a pre-coated microplate, lyophilized orexin standard, enzyme conjugate, wash concentrate, chromogenic TMB substrate, stop solution, and a protocol booklet for research use only.
