Human OPN/Osteopontin ELISA Kit

From Assay Genie, this colorimetric sandwich ELISA is designed for the quantitative measurement of human osteopontin (OPN) in plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Osteopontin is a multifunctional protein involved in bone remodeling, immune response modulation and tumor progression, and its overexpression has been linked to cancer, cardiovascular disorders and inflammatory conditions, making it a significant biomarker in disease research and therapeutic development.

Reactivity is human, and kit components are intended for storage at 2–8°C for 12 months. The workflow follows a six-step sandwich protocol: equilibrate reagents and TMB substrate to room temperature and load standards, samples and controls on the pre-coated plate; incubate at 37°C for 90 minutes; add biotin-labelled detection antibody and incubate at 37°C for 60 minutes; add HRP-Streptavidin (SABC) and incubate at 37°C for 30 minutes; develop with TMB substrate in the dark for 10–20 minutes; then add stop solution and read absorbance at 450 nm.

Reported performance includes EDTA plasma recovery of 85–100% (average 90%) and heparin plasma recovery of 86–102% (average 93%), with linearity for EDTA plasma of 87–100% at 1:4 and 80–94% at 1:8 dilution, and for heparin plasma of 87–105% (1:2), 83–101% (1:4) and 81–92% (1:8).
