Description

From Assay Genie, this Sandwich ELISA kit enables quantitative measurement of human Oncoprotein-Induced Transcript 3 (OIT3), a protein also referred to by the synonyms LZP and Liver-specific zona pellucida domain-containing protein (UniProt Q8WWZ8). The assay uses a double-antibody sandwich format with colorimetric detection, and is validated for use with human samples including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples.

Reported sensitivity is 0.188 ng/mL with a working range of 0.313–20 ng/mL. Reagents are stored at 2–8 °C for 12 months. Linearity has been demonstrated across 1:2, 1:4 and 1:8 dilutions: serum (n=5) 86–100%, 85–103%, 86–102%; EDTA plasma (n=5) 84–101%, 83–101%, 84–101%; heparin plasma (n=5) 81–100%, 82–97%, 80–100%. Recovery rates across the same matrices average 92% (serum, range 88–98%), 91% (EDTA plasma, range 85–95%) and 94% (heparin plasma, range 88–100%). The protocol involves equilibrating reagents and TMB substrate to room temperature, primary incubation at 37 °C for 90 minutes, biotin-labelled detection antibody incubation at 37 °C for 60 minutes, HRP-Streptavidin binding at 37 °C for 30 minutes, TMB colour development in the dark for 10–20 minutes, addition of stop solution and absorbance measurement at 450 nm.

OIT3 has been described as being intricately involved in tumour growth and progression, with its presence implicated in various cancer types, making the kit a useful tool for cancer-related biomarker research.
