From Assay Genie, this sandwich ELISA is intended for the quantitative measurement of human Nuclear receptor subfamily 1 group I member 2 (NR1I2), also referred to as Pregnane X receptor (PXR), a nuclear receptor central to xenobiotic and drug metabolism. The assay is reactive with human samples and validated for use with serum, plasma, tissue homogenates, cell culture supernates and other biological fluids, with specificity for natural and recombinant human NR1I2.

Reported performance characteristics include a detection range of 0.156-10ng/mL and a sensitivity of 0.045ng/mL, with intra-assay CV of 5.8% and inter-assay CV of 10.8%. Recovery is documented for serum (average 94%, range 88-100%) and plasma (average 96%, range 90-102%), and linearity has been assessed across serial dilutions in serum, EDTA plasma and heparin plasma matrices.

NR1I2 is a transcription factor that binds a response element in the promoters of CYP3A4 and ABCB1/MDR1, modulating genes involved in drug metabolism and detoxification, and is activated by compounds such as rifampicin as well as endogenous steroids such as pregnenolone and progesterone. The receptor functions as a heterodimer with RXR and interacts with NCOA1, and is localised to the nucleus. For research use only.
