Human NOTUM/Protein notum homolog ELISA Kit

Assay Genie provides a Sandwich ELISA Kit specifically designed for the precise detection of NOTUM (Protein notum homolog) protein levels in human samples. The assay uses a double-antibody sandwich format with colorimetric detection and is suitable for measuring NOTUM in serum, plasma, cell culture supernatants, cell or tissue lysates, and other liquid samples.

The kit offers a sensitivity of 46.875 pg/mL and a detection range of 78.125–5000 pg/mL. Reported linearity values (at 1:2, 1:4 and 1:8 dilutions) are 87–103%, 85–105% and 92–105% for serum (n = 5); 86–98%, 85–99% and 82–100% for EDTA plasma (n = 5); and 82–98%, 82–98% and 85–96% for heparin plasma (n = 5). Recovery values are 90–102% (average 96%) for serum, 89–100% (average 97%) for EDTA plasma, and 86–103% (average 91%) for heparin plasma. Reagents are stored at 2–8°C for up to 12 months.

NOTUM is a key enzyme that regulates the Wnt signaling pathway, which is involved in embryonic development, tissue homeostasis, and cancer progression. Dysregulation of NOTUM has been implicated in cancer and Alzheimer's disease, making it a relevant biomarker for studying these conditions. The assay workflow includes primary incubation at 37°C, detection antibody binding, HRP-Streptavidin binding, TMB color development in the dark, and absorbance measurement at 450 nm.
