Human NOTCH4 ELISA Kit

A sandwich ELISA manufactured by Assay Genie, designed for quantitative detection of human Notch4 using a double-antibody format with colorimetric readout. The assay is validated for use with human serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid sample types, and provides a tool for studying Notch4 in the Notch signaling pathway, where its dysregulation has been linked to cancer, cardiovascular and neurological conditions.

Reported sensitivity is 0.188 ng/mL with a working range of 0.313–20 ng/mL. Linearity for serum, EDTA plasma and heparin plasma at 1:2, 1:4 and 1:8 dilutions falls between 82% and 105%, and recovery averages 91% in serum, 96% in EDTA plasma and 94% in heparin plasma. Kits are stored at 2–8°C for 12 months.

The workflow includes equilibration of reagents and TMB substrate, loading of standards, samples and blanks, a 90-minute primary incubation at 37°C, a 60-minute biotinylated detection antibody step, a 30-minute HRP-Streptavidin (SABC) binding step, TMB colour development in the dark for 10–20 minutes, and immediate absorbance reading at 450 nm after stop solution.
