Human NOTCH3 ELISA Kit

Assay Genie provides a sandwich ELISA, double antibody platform for the quantitative measurement of human NOTCH3, also referenced by synonyms including CADASIL, CASIL, and neurogenic locus notch homolog protein 3. The kit is intended for research applications in developmental biology and is reactive with human samples. Notch 3 is a receptor protein implicated in cell fate determination, development, and the regulation of stem cell populations, and dysregulated signalling has been linked to cancer, cardiovascular disorders, and developmental abnormalities.

The assay accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Linearity in serum, EDTA plasma, and heparin plasma spans 80% to 103% across 1:2, 1:4, and 1:8 dilutions. Recovery is reported as 85-102% in serum (average 96%), 89-100% in EDTA plasma (average 94%), and 89-105% in heparin plasma (average 95%).

The procedure involves equilibrating reagents and TMB substrate to room temperature, a primary incubation at 37°C for 90 minutes, biotinylated detection antibody binding at 37°C for 60 minutes, HRP-Streptavidin incubation at 37°C for 30 minutes, TMB colour development in the dark for 10-20 minutes, and absorbance reading at 450 nm after the addition of stop solution. Unopened kit components should be stored at 2-8°C for 12 months.
