Description

Assay Genie offers a Sandwich ELISA designed for the quantitative measurement of human non-histone chromosomal protein HMG-14 (HMGN1) in serum, plasma, tissue homogenates, cell culture supernatants and other biological fluids, using a colorimetric detection method. The kit specifically recognises natural and recombinant human HMG-14, also known by the synonyms High mobility group nucleosome-binding domain-containing protein 1 and HMG14, with reported reactivity for human samples.

Performance characteristics taken from the manufacturer's product specification include a detection range of 0.312–20 ng/mL, a sensitivity of 0.19 ng/mL, an intra-assay CV of 4.6% and an inter-assay CV of 8.6%. Linearity has been evaluated in serum, EDTA plasma and heparin plasma across dilution factors of 1:2, 1:4, 1:8 and 1:16. Recovery is reported as an average of 94% (range 88–100%) for serum and 96% (range 90–102%) for plasma. Reagent components are supplied for storage at -20°C and 4°C, and the kit is intended for research use only.

HMGN1 is a non-histone chromosomal protein of the HMG-14/HMG-17 family (UniProt P05114) that binds to the inner side of nucleosomal DNA, modulating DNA–histone octamer interactions and helping to maintain transcribable genes in an open chromatin conformation. It also inhibits phosphorylation of nucleosomal histones H3 and H2A by RPS6KA5/MSK1 and RPS6KA3/RSK2. The protein is localised to the nucleus and cytoplasm, with cytoplasmic enrichment upon phosphorylation, and the gene is mapped to chromosomal location 21q22.2. The associated research area is Neurosciences.
