Description

A Sandwich ELISA from Assay Genie designed for the quantitative measurement of the human Nociceptin receptor (OPRL1) in serum, plasma, tissue homogenates, cell culture supernates and other biological fluids. The kit is intended for research use only and detects natural and recombinant human Nociceptin receptor, also known by the synonyms Kappa-type 3 opioid receptor, Orphanin FQ receptor, KOR-3, OOR and ORL1. The Uniprot accession for the target protein is P41146.

The assay provides a detection range of 78-5000pg/mL with a sensitivity of 27pg/mL. Reported precision values are an Intra-assay CV of 5.1% and an Inter-assay CV of 9.4%. Linearity has been evaluated for serum, EDTA plasma and heparin plasma across dilution factors of 1:2, 1:4, 1:8 and 1:16, with serum ranging from 99-119%, EDTA plasma from 87-117% and heparin plasma from 104-116%. Recovery in serum averaged 94% (range 88-100%) and in plasma averaged 96% (range 90-102%).

Nociceptin receptor is a G-protein coupled opioid receptor that functions as a receptor for the endogenous neuropeptide nociceptin. It is localized to the cell membrane as a multi-pass membrane protein and is also found in cytoplasmic vesicles following ligand-induced internalization, which requires phosphorylation at Ser-363. Binding of nociceptin triggers G-protein-mediated signaling that inhibits adenylate cyclase activity and calcium channel activity, while arrestins can activate MAP kinase pathways. The receptor plays a role in modulating nociception, perception of pain, and regulation of locomotor activity by the neuropeptide nociceptin.
