NFATC3 (Nuclear Factor of Activated T Cells 3), also known as NFAT4, is a calcium/calcineurin-regulated transcription factor of the NFAT family. In resting immune cells, NFATC3 resides in the cytoplasm in a phosphorylated, inactive state; upon intracellular calcium influx, the phosphatase calcineurin dephosphorylates NFATC3, triggering nuclear translocation and transcriptional activation of cytokine genes such as IL-2 and IL-4. This transcription factor is central to T-cell activation, immune tolerance, and adaptive immunity, and dysregulation of NFATC3 signaling has been investigated in transplant rejection, inflammatory disorders, cardiac hypertrophy, and tumor immunology.
