Human Neuropathy target esterase/PNPLA6 ELISA Kit

From Assay Genie, this Sandwich ELISA kit enables quantitative measurement of human Neuropathy target esterase (PNPLA6), also known as NTE or Patatin-like phospholipase domain-containing protein 6. The kit is intended for research use in the area of metabolism.

The kit is validated for human samples and accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reported sample preparation protocols include clotting and centrifugation of serum at 1000 × g for 20 minutes, anticoagulant collection of plasma followed by centrifugation at 1000 × g for 15 minutes at 2–8°C, homogenisation of tissue in PBS with protease inhibitors, centrifugation of cell culture supernatant at 2500 rpm for 5 minutes, and lysis of cells with protease inhibitors. Linearity has been demonstrated for serum, EDTA plasma and heparin plasma at 1:2, 1:4 and 1:8 dilutions, and recovery ranges are reported for the same matrices. Reagent components are intended to be stored at 2–8°C for up to 12 months, with TMB substrate and other light-sensitive reagents protected from direct light.

The assay procedure involves equilibrating reagents to room temperature, a primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody incubation at 37°C for 60 minutes, HRP-Streptavidin (SABC) binding at 37°C for 30 minutes, TMB colour development in the dark for 10–20 minutes, and a stop reaction with absorbance read at 450 nm. The kit is supplied with a pre-coated ELISA microplate, lyophilised standard, biotin-labelled detection antibody concentrate, HRP-Streptavidin conjugate concentrate, TMB substrate, sample dilution buffer, antibody dilution buffer, SABC dilution buffer, stop solution, wash buffer, plate sealers and a technical manual.
