Description

From Assay Genie, this sandwich ELISA, double antibody kit enables quantitative measurement of human Netrin-1 in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The colorimetric assay is reactive with human specimens, with absorbance read at 450 nm following TMB substrate development.

The kit has a stated sensitivity of 18.75 pg/mL and a standard range of 31.25–2000 pg/mL. Linearity has been demonstrated in serum (n=5) at 88–103% (1:2), 92–105% (1:4) and 89–105% (1:8); in EDTA plasma (n=5) at 90–98% (1:2), 85–91% (1:4) and 84–99% (1:8); and in heparin plasma (n=5) at 83–99% (1:2), 84–97% (1:4) and 81–95% (1:8). Recovery ranges were 93–105% (serum), 86–94% (EDTA plasma) and 85–99% (heparin plasma), with average recoveries of 99%, 91% and 93% respectively.

The procedure involves a 90-minute primary incubation at 37°C, a 60-minute biotin-labelled detection antibody step, 30 minutes of HRP-Streptavidin binding, and 10–20 minutes of TMB colour development in the dark. Reagents should be stored at 2–8°C for up to 12 months. Netrin-1 (Uniprot O95631; gene Ntn1) is implicated in axon guidance and neuronal development, with dysregulation associated with neurological disorders including Alzheimer's disease, Parkinson's disease and spinal cord injury, making this kit relevant to cell death research and the study of these conditions.
