Description

Assay Genie's Human Nestin ELISA Kit is a sandwich ELISA (double antibody) designed for the quantitative detection of Nestin in human samples. Compatible sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit uses a colorimetric detection method and is intended for use within neuroscience research applications.

Reported performance characteristics include a sensitivity of 9.375 pg/mL and an assay range of 15.625-1000 pg/mL. Linearity for serum samples (n = 5) is 83-98% (1:2), 88-97% (1:4), and 91-97% (1:8); for EDTA plasma (n = 5) it is 84-100%, 83-98%, and 81-98%; and for heparin plasma (n = 5) it is 85-99%, 81-96%, and 86-101%. Recovery ranges are 87-96% (average 91%) for serum (n = 5), 91-102% (average 95%) for EDTA plasma (n = 5), and 83-99% (average 95%) for heparin plasma (n = 5). Reagents should be stored at 2-8°C, where they remain stable for 12 months.

The procedure involves equilibrating reagents and TMB substrate to room temperature, a primary incubation at 37°C for 90 minutes, biotin-labeled detection antibody binding at 37°C for 60 minutes, HRP-Streptavidin (SABC) binding at 37°C for 30 minutes, TMB color development in the dark for 10–20 minutes, and absorbance measurement at 450 nm after adding stop solution. Nestin (synonyms: NES, FLJ21841; UniProt P48681) is described as a protein involved in regulating cellular structure and function, particularly in neuronal development and regeneration.
