Human Neprilysin-2 ELISA Kit

A colorimetric, sandwich double-antibody ELISA from Assay Genie enables quantitative measurement of human neprilysin-2. The kit is reactive with human samples and is suitable for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, with applications in cell biology research.

Manufacturer data report a sensitivity of 18.75 pg/mL and a standard range of 31.25-2000 pg/mL. Linearity was tested at 1:2, 1:4 and 1:8 dilutions (n=5): serum 85-100%, 87-105% and 94-102%; EDTA plasma 86-101%, 90-98% and 83-92%; heparin plasma 82-95%, 82-90% and 82-96%. Spike-and-recovery testing gave average recoveries of 96% in serum, 94% in EDTA plasma and 97% in heparin plasma. Unopened kit components are stored at 2-8°C for 12 months.

The assay workflow comprises a 90-minute primary incubation at 37°C, a 60-minute biotin-labelled detection antibody step, a 30-minute HRP-Streptavidin binding step, TMB development in the dark for 10-20 minutes and a stop reaction read at 450 nm. Neprilysin-2 (Uniprot Q495T6) is a metallo-endopeptidase whose dysregulation has been implicated in Alzheimer's disease, heart failure and diabetes through altered processing of substrates such as neuropeptides and amyloid beta.
