Description

Assay Genie's sandwich ELISA kit is designed for the quantitative measurement of human Necdin (NDN) in biological samples. The colorimetric assay is validated for use with serum, plasma, tissue homogenates, cell culture supernates and other biological fluids, and is intended for research use only. It offers specificity for natural and recombinant human Necdin and is suited to studies of cell growth, differentiation and apoptosis.

Reported performance characteristics include a detection range of 0.312–20 ng/mL and a sensitivity of 0.1 ng/mL. Precision is documented with an intra-assay CV of 4.5% and an inter-assay CV of 9.8%. Linearity has been assessed across serum, EDTA plasma and heparin plasma at 1:2, 1:4, 1:8 and 1:16 dilutions: serum spanned 94–102%, 93–103%, 89–99% and 105–116%; EDTA plasma spanned 110–120%, 114–124%, 84–96% and 91–101%; heparin plasma spanned 109–119%, 96–106%, 100–113% and 105–115%. Recovery testing shows serum averaging 98% (range 92–104%) and plasma averaging 100% (range 94–106%).

Necdin functions as a growth suppressor that facilitates entry of the cell into cell cycle arrest and behaves similarly to retinoblastoma protein, repressing cell-cycle-promoting proteins such as SV40 large T antigen, adenovirus E1A and the transcription factor E2F. It interacts with p53 in an additive manner to inhibit cell growth and additionally acts as a transcription factor that binds guanosine-rich DNA sequences. Dysregulation of Necdin is associated with Prader-Willi syndrome and cancer, supporting its relevance as a biomarker for disease pathogenesis research.
