Human NDST2 ELISA Kit

From Assay Genie, this Sandwich ELISA Kit uses a double-antibody format for the quantitative detection of NDST2 in human samples. The kit accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, and uses a colorimetric read-out with absorbance measured at 450 nm following TMB substrate development. Reagents are stored at 2-8°C for up to 12 months.

Reported performance characteristics include a sensitivity of 46.875 pg/mL and an assay range of 78.125–5000 pg/mL. Linearity across 1:2, 1:4 and 1:8 dilutions was 86–105%, 84–101% and 82–97% for serum (n=5); 86–102%, 83–92% and 81–100% for EDTA plasma (n=5); and 86–104%, 87–100% and 80–100% for heparin plasma (n=5). Spike-and-recovery averaged 97% for serum (range 91–102%, n=5), 95% for EDTA plasma (range 86–102%, n=5) and 93% for heparin plasma (range 87–100%, n=5).

NDST2 (Uniprot P52849) is a bifunctional enzyme that modifies heparan sulfate through N-deacetylation and N-sulfotransferase activity, and is relevant to studies of cell signalling, cancer and metabolic disorders. The protocol follows a standard sandwich workflow: equilibrate reagents and TMB substrate, incubate standards and samples at 37°C for 90 minutes, add biotin-labelled detection antibody for 60 minutes, incubate with HRP-Streptavidin (SABC) for 30 minutes, develop with TMB in the dark for 10–20 minutes, then add stop solution and read at 450 nm.
