Description

Designed for the quantitative measurement of human Necdin (NDN), this colorimetric immunoassay from Assay Genie is configured as a sandwich ELISA using a double-antibody format and is reactive with human samples. The kit has been validated for use with serum and plasma sample types and is intended for research applications in developmental biology.

Necdin is described in the supporting documentation as a growth suppressor that facilitates entry of the cell into cell cycle arrest, functioning similarly to the retinoblastoma protein by binding to and repressing cell-cycle-promoting proteins such as SV40 large T antigen, adenovirus E1A, and the transcription factor E2F. The protein also interacts with p53 in an additive manner to inhibit cell growth and acts as a transcription factor that binds directly to specific guanosine-rich DNA sequences. The NDN gene is located in the Prader-Willi syndrome deletion region on chromosome 15q11.2-q12, is an intronless imprinted gene expressed exclusively from the paternal allele, and maps to the UniProt entry Q99608. Dysregulation of Necdin expression has been linked to Prader-Willi syndrome and to cancer, making the protein a relevant biomarker for research into these conditions.

According to the manufacturer, the assay has a stated sensitivity of 0.188 ng/mL and a working range of 0.313–20 ng/mL, and storage is recommended at 2–8°C for 12 months. Reported linearity values for serum, EDTA plasma, and heparin plasma are provided across dilution factors, together with recovery ranges in serum, EDTA plasma, and heparin plasma. Sample preparation guidance covers serum, plasma, tissue homogenate, cell culture supernatant, and cell lysate workflows.
