NADH dehydrogenase subunit 1 (ND1), a hydrophobic core subunit of mitochondrial respiratory complex I encoded by the MT-ND1 gene, can be quantified in human research samples using this Assay Genie ELISA Kit. ND1 sits at the matrix interface of complex I and couples electron transfer from NADH to ubiquinone during oxidative phosphorylation, so changes in its abundance serve as a biomarker in studies of mitochondrial bioenergetics, reactive oxygen species generation, and ATP turnover in cell culture and tissue models.

The kit is supplied as a complete package containing a pre-coated microplate, lyophilised recombinant ND1 standards, biotinylated detection antibody, streptavidin-HRP conjugate, wash concentrate, TMB substrate, and stop solution, each in a dedicated vial and sufficient material for one full microplate run of unknowns in duplicate alongside an eight-point standard curve. Researchers apply it to investigate complex I content in models of neurodegeneration, ischaemia–reperfusion injury, metabolic disease, and inherited mitochondrial disorders, and to monitor ND1 expression following pharmacological knockdown, CRISPR editing, or exercise interventions.

The protocol follows a standard capture-phase incubation, wash, detection-antibody binding, and signal-development workflow typical of plate-based immunoassays performed for research use only, and the manual provides recommended starting dilutions for serum-derived and plasma-derived samples. Each kit supports one plate, and unopened components should be stored refrigerated in the dark until the printed expiration date; reconstituted reagents should be aliquoted and frozen to limit repeated freeze-thaw cycles.
