Nicastrin (NCSTN) is the substrate-recognition subunit of the gamma-secretase intramembrane protease complex, which cleaves amyloid precursor protein and Notch receptors and is a focal point in Alzheimer's disease research. Loss-of-function mutations in NCSTN are also linked to familial hidradenitis suppurativa, broadening the kit's relevance across neuroscience and dermatology laboratories.

This colorimetric ELISA from Assay Genie is configured to quantify Nicastrin concentrations in human-derived matrices, with all critical reagents supplied ready to use: a pre-coated microplate, biotinylated detection antibody, streptavidin-HRP conjugate, TMB substrate, calibrated standards, wash buffer, and stop solution. The protocol supports trained researchers running quantitative immunoassays on a range of human specimens including cell culture supernatant and tissue homogenate, and the assay is intended for research use only.

After incubation and substrate development, absorbance is read on a standard plate reader and Nicastrin concentration is interpolated from a freshly generated standard curve. Unused strip wells should be resealed with the desiccant pack and stored refrigerated, while all reagents should be equilibrated to room temperature before each run to ensure consistent binding kinetics and reproducible results. Scientists studying gamma-secretase activity, APP processing dynamics, or NCSTN-associated skin disorders will find this kit useful as a quantitative biomarker tool.
