The double-antibody, colourimetric format delivers a sensitivity of 9.375 pg/mL across a working range of 15.625-1000 pg/mL, and the assay is validated for serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, with reagents stable at 2-8°C for 12 months.

Validation data show consistent performance across matrices. Linearity in serum (n=5) ranged from 92-105% at 1:2, 92-100% at 1:4, and 91-100% at 1:8; EDTA plasma (n=5) gave 89-95%, 85-101%, and 91-100% at the same dilutions, while heparin plasma (n=5) gave 91-105%, 85-98%, and 84-101%. Spike-and-recovery averaged 94% in serum, 94% in EDTA plasma, and 91% in heparin plasma, with corresponding recovery ranges of 86-104%, 85-99%, and 86-100%. The procedure involves equilibrating reagents and the TMB substrate to room temperature, loading standards and samples for primary incubation at 37°C for 90 minutes, incubation with biotin-labelled detection antibody at 37°C for 60 minutes, HRP-Streptavidin binding at 37°C for 30 minutes, colour development with TMB substrate in the dark for 10-20 minutes, and a stop reaction with absorbance measured at 450 nm.
