Description

From Assay Genie, this sandwich ELISA uses a double-antibody format with colorimetric detection to quantify Alpha-N-Acetylglucosaminidase (NAG) in human samples, including serum, plasma, cell culture supernatant, cell or tissue lysates, and other liquid samples. NAG is an enzyme involved in the breakdown of complex carbohydrates, and altered levels have been associated with kidney disease, lysosomal storage disorders, and rheumatoid arthritis, making it a valuable biomarker for studying these conditions.

Performance data include a sensitivity of 0.188 ng/mL and a working range of 0.313–20 ng/mL. Recovery in spiked serum was 86–104% (average 99%), 86–104% in EDTA plasma (average 96%), and 88–102% in heparin plasma (average 93%). Linearity across 1:2, 1:4 and 1:8 dilutions ranged 81–105% for serum, 84–101% for EDTA plasma, and 82–103% for heparin plasma. Kit components are stored at 2–8°C, with the microplate strips and lyophilized standard also stable for 12 months at –20°C.

The assay procedure involves equilibrating reagents and the TMB substrate to room temperature, loading standards and samples, incubation at 37°C (90 minutes for antigen binding, 60 minutes with biotinylated detection antibody, 30 minutes with HRP-Streptavidin), TMB substrate development in the dark for 10–20 minutes, and absorbance measurement at 450 nm after adding stop solution.
