NQO1, also called DT-diaphorase, is a cytosolic flavoenzyme that catalyzes the two-electron reduction of quinones to hydroquinones using NADH or NADPH as the electron donor. Because the enzyme sits at the intersection of xenobiotic detoxification, oxidative stress response, and bioactivation of quinone-based chemotherapeutics such as mitomycin C, NQO1 is routinely quantified in tumor biology, pharmacology, and environmental toxicology studies. The Assay Genie kit targets the human protein and is supplied as a complete set of reagents in a 96-assay configuration for research use only.

Sample inputs include tissue homogenates and cell lysates, covering most in vitro and ex vivo workflows. Investigators working on non-small cell lung cancer, where NQO1 is frequently overexpressed and linked to poor prognosis, use the assay to compare protein levels across primary tumors, adjacent normal tissue, and patient-derived xenografts. Toxicology groups focused on benzene, polycyclic aromatic hydrocarbons, and other quinone-generating exposures apply the kit to evaluate enzyme induction in hepatic and pulmonary cell cultures exposed to candidate pollutants.

Detection relies on a chromogenic method read on standard plate readers, with calibration standards included to interpolate unknown concentrations. The microplate-based format supports batch processing of dozens of samples in parallel, and researchers are advised to aliquot standards after first use to limit freeze-thaw cycles. Reconstituted reagents should be refrigerated when not in use to maintain assay performance.
