Description

Assay Genie offers a Sandwich ELISA for the quantitative measurement of human NAD-dependent protein deacetylase sirtuin-7 (SIRT7). The kit is designed for the accurate detection of SIRT7 in serum, plasma, tissue homogenates, cell culture supernates and other biological fluids, with reactivity confirmed for human samples. It is intended for research use only and supports work in the epigenetics research area.

Performance characteristics include a detection range of 0.312–20ng/mL and a sensitivity of 0.1ng/mL, with an intra-assay CV of 5.9% and inter-assay CV of 9.6%. The assay shows specificity for natural and recombinant human NAD-dependent protein deacetylase sirtuin-7. Linearity has been demonstrated in serum (N=5) at 1:2 to 1:16 dilutions, with recoveries ranging from 109% (serum, range 103–115%) and 111% (plasma, range 105–117%).

SIRT7 is a member of the sirtuin family of proteins, acting as an NAD-dependent protein deacetylase that specifically mediates deacetylation of histone H3 at 'Lys-18' (H3K18Ac), a mark linked to control of gene expression. The protein is associated with the nucleolus, nucleus and cytoplasm, and is involved in processes including rRNA transcription, DNA repair, and cellular metabolism, with dysregulation linked to cancer, metabolic disorders, and aging. Synonyms include Regulatory protein SIR2 homolog 7, SIR2-like protein 7, and SIR2L7, with UniProt entry Q9NRC8.
