NAALAD2, also known as glutamate carboxypeptidase III (GCPIII), is a membrane-anchored zinc metallopeptidase (EC 3.4.17.21) that hydrolyses the neuropeptide N-acetyl-L-aspartyl-L-glutamate (NAAG) into N-acetylaspartate and free glutamate. Expressed in testis, kidney and selected regions of the nervous system, NAALAD2 helps regulate extracellular glutamate signalling and contributes to folate polyglutamate catabolism, making it a relevant target in neuroscience, neurology and prostate carcinoma research programmes.

Built by Assay Genie around matched monoclonal antibody pairs, the immunoassay uses pre-coated microplate wells, a calibrated recombinant NAALAD2 standard, biotinylated detection antibody, streptavidin-HRP conjugate, wash buffer, chromogenic substrate and stop solution. Researchers pipette calibrators and diluted research specimens into the wells, incubate, wash and develop the chromogen for absorbance readout on a standard plate reader. Reconstituted standards and concentrated detection antibody should be aliquoted and frozen, while working dilutions may be kept refrigerated. Typical incubation temperatures are 37°C during the binding step and room temperature during substrate development. The kit contains all reagents required for one plate of determinations and is intended for in vitro research use only, not for clinical diagnostic procedures.
