ELISA-format immunoassay from Assay Genie for quantitative measurement of human N-Acetylgalactosaminidase Alpha (NAGa, also called α-GalNAcase), the lysosomal exoglycosidase encoded by the NAGA gene. NAGa hydrolyzes terminal α-linked N-acetylgalactosamine residues from glycoproteins, glycopeptides, and glycolipids, and its deficiency underlies two rare autosomal-recessive lysosomal storage conditions: Schindler disease (types I, II, and III) and Kanzaki disease. Reliable quantification of the enzyme in patient and model-system specimens therefore supports diagnostic biomarker research, enzyme replacement studies, and characterization of residual activity in fibroblasts and other tissues.

The 96-assay kit is supplied as a complete set of reagents including a pre-coated microplate, recombinant standards, biotinylated detection antibody, streptavidin-HRP conjugate, TMB substrate, wash buffer, and stop solution, sufficient for running duplicate standard curves and unknowns on a single plate. It is reactive with the human enzyme and intended for use with processed human specimens. The workflow follows a conventional capture-analyte-detection sequence with a TMB-based absorbance endpoint read on a standard microplate reader.

Typical research applications include profiling NAGa levels across patient sample matrices, comparing enzyme concentrations in tissue homogenates and cell lysates from healthy versus affected donors, monitoring expression changes in cell-line models of NAGA deficiency, and supporting compound-screening or genetic-complementation studies aimed at restoring α-GalNAcase activity. The kit format is compatible with standard batch processing workflows and integrates with routine immunology and cell-biology laboratory pipelines.

Store kit components according to the enclosed product insert; reagents are kept refrigerated between uses, with longer-term storage directions provided for individual components. Bring all reagents to ambient temperature before initiating the assay and avoid repeated freeze-thaw cycles of diluted working solutions. Protect light-sensitive substrates from extended exposure during incubation steps.
