Description

Assay Genie supplies a Sandwich ELISA designed for the quantitative measurement of human Myosin light chain kinase, smooth muscle (MYLK) in research samples. The kit is validated for use with serum, plasma, tissue homogenates, cell culture supernates and other biological fluids and is intended for research use only. The target protein corresponds to UniProt entry Q15746 and is also known by the synonyms Kinase-related protein, Telokin, KRP, MLCK, MLCK1 and MYLK1.

Performance characteristics include a detection range of 0.156-10ng/mL and a sensitivity of 0.076ng/mL. Precision is reported with an intra-assay CV of 5.2% and an inter-assay CV of 7.9%. Linearity has been examined in serum, EDTA plasma and heparin plasma at dilutions of 1:2, 1:4, 1:8 and 1:16. Recovery averages 103% (range 97-109%) in serum and 105% (range 99-111%) in plasma. The kit falls within the Signal Transduction research area.

Myosin light chain kinase (smooth muscle) is a calcium/calmodulin-dependent enzyme implicated in smooth muscle contraction via phosphorylation of myosin light chains (MLC). It also regulates actin-myosin interaction through a non-kinase activity and phosphorylates PTK2B/PYK2 and myosin light chains. The protein is localised to the cytoplasm, cell projections, lamellipodium, cleavage furrow and cytoskeleton, and is reported at stress fibers during interphase and at the cleavage furrow during mitosis.
