Description

This Sandwich ELISA from Assay Genie is specifically designed for the precise measurement of myogenin levels in human samples including serum, plasma, and cell culture supernatants. Using a double antibody format, the assay is described as offering high sensitivity and specificity, delivering dependable and consistent results suited to a variety of research purposes. Myogenin is a key protein that plays a critical role in the differentiation of muscle cells, making it an important biomarker for studying muscle development and regeneration, as abnormal myogenin levels have been linked to various muscle-related disorders and diseases.

The kit achieves a sensitivity of 0.375 ng/mL across a detection range of 0.625-40 ng/mL and accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Storage is at 2-8°C for 12 months. Validation data show consistent linearity across matrices: at the 1:2 dilution, serum (n=5) recovers at 86-104%, EDTA plasma (n=5) at 82-100%, and heparin plasma (n=5) at 90-98%, with comparable performance at 1:4 and 1:8. Recovery averages are 93% for serum (range 87-104%), 97% for EDTA plasma (range 92-105%), and 100% for heparin plasma (range 90-105%).

The protocol follows a standard sandwich workflow: reagent and plate preparation, primary incubation at 37°C for 90 minutes, biotin-labeled detection antibody binding for 60 minutes at 37°C, HRP-Streptavidin binding for 30 minutes, TMB substrate development in the dark for 10-20 minutes, and absorbance reading at 450 nm after addition of stop solution. The target is listed under UniProt P15173 within the cell cycle research area, with synonyms including MYOG, Myogenin, BHLHC3, MYF4, bHLHc3, Myogenic factor-4, Class C basic helix-loop-helix protein 3, myf-4, Myf-4, MYF4myogenin, and Myogenic factor 4.
